αヘリックスの形成速度は-50℃でもミリ秒より速い

蛋白質のフォールディング初期過程にαヘリックスが形成される速度について、変性剤希釈法でミリ秒より遅いとのデータが発表されていた。我々は、16個と17個のオリゴペプチドを用いて、-50℃でもαヘリックスが形成される速度はミリ秒より速いと結論づけた。この結果は、温度ジャンプ法などの結果とよく一致しており、αヘリックスの形成がフォールディングの初期過程に重要な役割を果たす可能性を示唆した。

Formation rate of alpha-helix is faster than a few milliseconds even at -50℃

Previously, Clarke reported that alpha-helix formation rate is so slow as in millisecond region by means of denaturant-dilution jump. We repeated the same kind of experiments with 16th and 17th oligopeptides. Results we obtained demonstrated that all helical formation rate is faster than a few milliseconds even at -50℃. The results are very consistent with T-jump study so far reported, and it is suggested that alpha-formation might be an important role in the initial process of protein folding.
